Madera Biosciences is developing small molecule programs to increase the levels of functional, lipidated ApoE in the brain as therapeutics for early stages of Alzheimer's. They are developing molecules that are administered orally and which penetrate the brain to enhance the clearance of Aβ and interrupt the neuroinflammatory-Aβ cycle. Their 1st lead candidate is a small molecule, MAD8100, that works via an unknown target to increase ApoE levels by human brain astrocytes. The 2nd lead candidate, MAD7001, binds to the nuclear receptor RXR and works by increasing ApoE release by human brain astrocytes.
